Clone REA467 recognizes the human G-protein coupled receptor 56 (GPR56) antigen, a multi-pass membrane protein, which is also known as TM7XN1. GPR56 is a member of the adhesion-GPCR family of receptors, which are characterized by an extended extracellular region with a variable number of protein domains coupled to a TM7 domain via a domain known as the GPCR-autoproteolysis inducing (GAIN) domain. It is expressed in a wide range of tissues, such as brain, thyroid gland, and heart. GPR56 has been shown to have numerous roles in cell guidance/adhesion as exemplified by its roles in tumor inhibition and neuron development. It is a receptor for collagen III/COL3A1 in the developing brain and involved in regulation of cortical development, specifically in maintenance of the pial basemant membrane integrity and in cortical lamination. Binding to the COL3A1 ligand inhibits neuronal migration and activates the RhoA pathway by coupling to GNA13 and possibly GNA12. More recently it has been shown to be a marker for cytotoxic T cells and a subgroup of natural killer cells. In melanocytic cells GPR56 gene expression may be regulated by MITF.
