Clone REA681 recognizes Syk protein phosphorylated at tyrosine 348. Syk is a 72 kDa non–receptor tyrosine kinase cytoplasmic protein. Syk belongs to the SYK/ZAP 70 family of tyrosine protein kinases and is mainly expressed in hematopoietic cells. Activated immunoreceptors, phosphorylated at ITAM motifs recruit Syk from the cytoplasm to the signalosome complex at the perimembrane location. Syk, upon binding of SH2 domains to immunoreceptor ITAM is activated and undergoes autophosphorylation and phosphorylation by SRC kinases. Activated Syk catalyzes tyrosine phosphorylation of multiple downstream proteins which regulate signaling pathways such AKT, MAP kinase, and NF-κB. A wide variety of cellular and physiological functions such as cell activation and differentiation are regulated by Syk. Phosphorylation at Tyr-348 creates a binding site for VAV1.
