Thrombopoietin (TPO), also termed megakaryocyte growth and development factor (MGDF), is a glycoprotein hormone and the major stimulator of megakaryopoiesis and platelet production. It promotes the proliferation of hematopoietic stem cells, primitive progenitors, megakaryocytes, and platelets.
MACS GMP Recombinant Human TPO is designed for ex vivo cell culture processing. No animal- or human-derived materials were used for the manufacture of this product. The product is lyophilized without carrier protein or preservatives.

Biological activity
TPO is expressed in liver, kidney, spleen, lung, bone marrow, and brain. The TPO receptor is a product of the proto-oncogene c-mpl and displays homology with type I cytokine receptor superfamily members. The receptor is present mainly on hematopoietic stem cells, megakaryocytic progenitors, megakaryocytes, and platelets.
MACS GMP Recombinant Human TPO can be used for a variety of applications, including the expansion2,3 and the viral transduction4,5 of hematopoietic stem and progenitor cells derived from bone marrow, peripheral blood cells, and cord blood, or the in vitro differentiation of megakaryocytes6.
Technical specifications
- Identity/ Molecular mass: 18804 Da determined by mass spectrometry. This corresponds to the first 174 amino acids of the mature human TPO including an N-terminal methionine.
- Purity: ≥ 85 % as determined by chip electrophoresis.
- Endotoxin content: < 50 EU/mg, as determined by kinetic Limulus Amoebocyte Lysate (LAL) assay (Pharmacopoeia Europaea (Ph. Eur.)).
- Residual host cell DNA content: < 20 ng/mg, as determined by quantitative PCR specific for E. coli genomic DNA.
- Residual host cell protein content: < 0.5 µg/mg, as determined by E. coli HCP ELISA.
