Interleukin 4 (IL-4) is an anti-inflammatory cytokine and is a key regulator in humoral and adaptive immunity. It has complex biological roles and promotes growth and differentiation of B cells and T cells, and cells of the monocytic lineage.

MACS GMP Recombinant Human IL-4 is designed for ex vivo cell culture processing. No animal- or human-derived materials were used for the manufacture of this product, unless otherwise stated in the respective Certificate of Origin. The product is lyophilized without carrier protein or preservatives.

Biological activity

Background information

IL-4 is predominantly secreted by activated CD4⁺ memory and effector T_H2 cells, basophils, and mast cells. It promotes the proliferation and differentiation of B cells, as well as immunoglobulin isotype switching, and MHC class II antigen and low-affinity IgE receptor expression. Furthermore, IL-4 induces the differentiation of naive CD4⁺ T cells into helper T_H2 cells, while suppressing T_H1 development, and promotes chemotaxis of mast cells and basophils.

Applications

MACS GMP Recombinant Human IL-4 can be used for a variety of applications, including the ex vivo generation of human dendritic cells from enriched CD14⁺ monocyte populations.^2–⁴

Identity/ Molecular mass: 15088 Da, determined by mass spectrometry. This corresponds to the mature form of human IL-4 including an N-terminal methionine (130 amino acids). Source E. coli.
Purity: ≥ 97 % as determined by chip electrophoresis.
Endotoxin content: < 50 EU/mg, as determined by kinetic Limulus Amoebocyte Lysate (LAL) assay (Pharmacopoeia Europaea (Ph. Eur.)).
Residual host cell DNA content: < 20 ng/mg, as determined by quantitative PCR specific for E. coli genomic DNA.
Residual host cell protein content: < 0.5 µg/mg, as determined by E. coli HCP ELISA.
