Recombinant Protein G (rProtein G) Agarose is a ready-to-use affinity chromatography matrix supplied as a 50% slurry (in phosphate buffered saline (PBS), pH 7.0; 20% ethanol). It is used for the purification of IgG from most mammalian species (see table), and does not bind IgM, IgA, IgE, or serum albumin. The rProtein G is a recombinant form of streptococcal protein G engineered to eliminate nonspecific binding to serum albumin, and is immobilized on 4% agarose beads. rProtein G Agarose has a binding capacity of approximately 20 mg of human IgG per mL of beads. rProtein G Agarose binds to a broader range of IgG isotypes than react with protein A. The binding characteristics and stability of rProtein G Agarose facilitate rapid and repeated use for affinity chromatography.
Using rProtein G Agarose
To purify IgG, load crude samples (serum, ascites fluid, or hybridoma supernatant) onto a column of rProtein G Agarose under physiologic conditions of salt and pH. Non-IgG components do not bind to the beads and elute in the flow through fraction. Purified IgG is then eluted with a low pH buffer.
