Clone REA412 recognizes the human lymphotoxin beta receptor (LT-ßR) antigen, a 47 kDa single-pass type I membrane protein which is also known as tumor necrosis factor receptor superfamily member 3 (TNF-RIII). LT-ßR is a member of the TNF receptor family that play a role in the development and organization of lymphoid tissues. Unlike the other TNF receptors, LT-ßR does not bind a soluble ligand, but rather, a membrane form of a heterotrimeric lymphotoxin complex comprised of one LTα and two LTβ molecules. The development of lymph nodes is a process that involves the LT-ßR dependent crosstalk between mesenchymal lymphoid tissue organizer and hematopoietic lymphoid tissue inducer cells. Cell death induced by LT-ßR is dependent on TRAF3, whereas NFκB activation appears to be mediated by TRAF5 or TRAF2. LT-ßR is expressed on a variety of human primary and transformed cells, but is absent on T or B lymphocytes and primary monocytes or peripheral dendritic cells, although LT-ßR is detected on some myeloid leukemic lines. In the developing thymus LT-ßR is prominent along the trabeculae and into the medulla up to corticomedullary junction. In the spleen, LT-ßR is prominently expressed by cells in the red pulp and along the borders of red and white pulp which colocalizes with reticular stromal cells.
