Stimulate T cells reactive to the Ag85B antigen of Mycobacterium tuberculosis using PepTivator® Peptide Pools. PepTivators are pools of lyophilized peptides, consisting mainly of 15-mer sequences with 11 amino acids overlap. PepTivator M. tuberculosis Ag85B is a peptide pool covering the complete sequence of the Ag85B protein (UniProtKB Acc. no. P9WQP0).
In vitro stimulation of antigen-specific T cells with PepTivator Peptide Pools causes the secretion of effector cytokines and the up-regulation of activation markers, which then allow the detection and isolation of antigen-specific T cells.
Tuberculosis (TB) caused by infection with Mycobacterium tuberculosis is one of the leading causes of death worldwide. According to the WHO a quarter of the world’s population is infected with M. tuberculosis. While most infections are latent and asymptomatic, about 5–10% of infections progress to active disease. TB persists as a serious global health threat, requiring new vaccines and therapeutics to combat multidrug resistance. The secreted protein antigen 85B (Ag85B) is the most highly expressed protein by M. tuberculosis. As a mycolyltransferase, Ag85B is involved in synthesis and maintenance of the bacterial cell wall. Ag85B is strongly immunogenic and has been found to equally induce humoral and cellular immunity. Due to its key role in mycobacterial physiology, Ag85B has emerged as a promising drug target and has also been studied as a TB vaccine component.
