Clone REA254 recognizes the CD137L antigen, a type II transmembrane glycoprotein belonging to the TNF superfamily, which is also known as 4-1BB ligand or tumor necrosis factor ligand superfamily member 9. CD137L is expressed on a variety of antigen-presenting cells, including dendritic cells (DCs), B cells, and macrophages as well as on activated T Lymphocytes. CD137L cross-links its receptor, CD137, which is expressed on activated T cells, and co-stimulates T cell activities. Signaling through interaction with CD137 by soluble CD137L has been shown to cause T cell–expansion, cytokine induction, and upregulation of antiapoptotic genes, and to prevent activation-induced cell death. In monocytes, macrophages, and DCs, CD137L signaling induces activation, migration, survival, and differentiation. The role of CD137 signaling has been studied in several autoimmune processes, including rheumatoid arthritis, experimental autoimmune encephalomyelitis, and systemic lupus erythematosus.
