Clone REA447 recognizes the human CD305 antigen, a single-pass type I membrane protein which is also known as leukocyte-associated immunoglobulin-like receptor 1 (LAIR-1). CD305 contains a single extracellular immunoglobulin-like domain and a cytoplasmic tail containing two immune receptor tyrosine-based inhibitory motifs. It is structurally related to several other inhibitory Ig superfamily members. CD305 is expressed on almost all immune cells, including NK cells, T cells, B cells, monocytes, monocyte-derived dendritic cells, eosinophils, basophils, and mast cells. Besides being expressed on differentiated peripheral blood cells, the receptor is also present on CD34+ hematopoietic progenitor cells and on the majority of thymocytes. Cross-linking of CD305 on human NK cells delivers a potent inhibitory signal that is capable of inhibiting target cell lysis by resting and activated NK cells. Furthermore, CD305 can inhibit the cytotoxic activity of effector T cells upon CD3 cross-linking or antigen stimulation. In primary B cells, CD305 cross-linking leads to decreased BCR-induced calcium mobilization and down-regulation of Ig and cytokine production.
